Chromosome 17 variants influence prostate cancer, type 2 diabetes risk

NEW YORK (Reuters Health) - Two variants in chromosome 17 influence prostate cancer, and one of these also influences the risk for type 2 diabetes, according to a report in the July 1st advance online publication of Nature Genetics.

The long arm of chromosome 17 has previously been associated with prostate cancer in family-based linkage studies, the authors explain, but no susceptibility variants have been reported before now.

Dr. Kari Stefansson, from deCODE Genetics, Reykjavik, Iceland, and colleagues analyzed six single nucleotide polymorphisms (SNPs) on chromosome 17q that showed some association with prostate cancer in their genome-wide analysis.

Two SNPs showed significant association with prostate cancer, with allele A of rs4430796 conferring a 22% increased risk and allele G of rs1859962 conferring a 20% increased risk.

SNP rs4430796 is located in the second intron of the TCF2 gene, the authors report, whereas rs1859962 is in a gene-poor area on 17q24.3.

Allele A of SNP rs4430796 also showed a protective effect against type 2 diabetes, the researchers note, affording a 14% reduced risk.

SNP rs4430796 was substantially correlated with another SNP in the first intron of TCF2 (rs7501939), whose allele C conferred a similar risk increase for prostate cancer and protection against type 2 diabetes.

"Together, based on the combined results, these two variants have an estimated joint population attributable risk of about 36%, which is substantial from a public health viewpoint," the investigators write.

"We favor the notion that the primary functional impact of rs4430796 (or a presently unknown correlated variant) is on one or more metabolic or hormonal pathways important for the normal functioning of individuals throughout their lives that incidentally modulate the risk of developing prostate cancer and type 2 diabetes late in life," the researchers note.
