Proepithelin Key to Bladder Cancer Spread; Potential Drug Target
NEW YORK (Reuters Health) Jul 26 - The growth factor proepithelin induces migration and invasion of bladder cancer, two key steps leading to metastasis, scientists report in the July 15th issue of Cancer Research.
"Although bladder cancer is one of the most common malignancies, very little is known about the molecular mechanisms that determine malignant transformation in the urothelial lining the bladder wall," Dr. Andrea Morrione and colleagues from the Kimmel Cancer Center, Thomas Jefferson University in Philadelphia point out in the paper.
Most bladder tumors arise in the bladder epithelium, and often recur and/or metastasize, regardless of treatment with surgery, chemotherapy, or immunotherapy, they also note.
"Our results," write Dr. Morrione and colleagues, "provide the first evidence for a role of proepithelin in stimulating migration and invasion of bladder cancer cells, and support the hypothesis that this growth factor may play a critical role in the establishment of the invasive phenotype."
Proepithelin, which is also known as progranulin, acrogranin, PC-derived growth factor, or granulin-epithelin precursor, is an important regulator of cell growth, migration, and transformation. The growth factor is overexpressed in breast, ovarian, and renal cancer as well as in glioblastomas.
Dr. Morrione and colleagues investigated the effects of proepithelin on transitional cell carcinoma-derived cells using purified human recombinant proepithelin.
Although proepithelin did not appreciably affect cell growth, it did enhance motility and invasion of the cells, as mentioned. It also stimulated in vitro wound closure, as has been shown in previous studies.
These effects required the activation of the MAPK pathway and paxillin, a protein associated with cellular attachment.
With continued study, proepithelin may turn out to be a useful clinical marker for diagnosis and prognosis in bladder tumors, conclude Dr. Morrione and colleagues. It could also provide new leads for improved therapeutic approaches against this cancer. [标签:content1][标签:content2]
