NEW YORK (Reuters Health), Dec 4 - The heightened risk for leukemia in children with Down syndrome does not appear to be related to ionizing radiation exposure associated with medical testing, researchers with the Children's Oncology Group report in the November issue of Pediatrics
The incidence of leukemia among Down syndrome children is 10- to 20-fold higher than in children without Down syndrome.
"Leukemia in children with Down syndrome is likely multifactorial and it is thought that the trisomy is likely the first in a series of genetic 'hits,'" Dr. Julie A. Ross and Amy M. Linabery of the University of Minnesota, Minneapolis, told Reuters Health. "The current study was undertaken as it was hypothesized that x-ray irradiation may be one of these environmental hits."
The Children's Oncology Group studied 158 children with Down syndrome and leukemia and 173 children with Down syndrome without leukemia, who served as controls. Of the cases, 97 children had acute lymphoblastic leukemia and 61 children had acute myeloid leukemia.
Radiation exposure before conception, in utero and postnatally was assessed.
Dr. Ross and Linabery told Reuters Health that "this study failed to find a moderate to large effect of diagnostic x-ray irradiation in the etiology of leukemia among children with Down syndrome."
"The lack of a strong association between postnatal medical test exposure and acute leukemia among children with Down syndrome should be reassuring for physicians, as well as concerned parents, as these children are subjected to diagnostic irradiation in the course of their standard primary care," the investigators commented.
"Caution should be exercised in interpreting these results, as this is the first study to investigate this association among this patient population," they advised. "Future studies should include greater sample sizes as well as more precise methods of dose evaluation."
