【drug-news】动物实验显示:ARB类药物可增加骨骼

NEW YORK (Reuters Health) Sept 18 - In an animal model of acute muscle injury, treatment with an angiotensin II receptor blocker (AR enhances muscle regeneration and decreases fibrosis, according to a report in the August issue of The American Journal of Sports Medicine.

In a previous study, patients treated with angiotensin-modulating medications for hypertension showed the unexpected side effect of decreased rates of muscle wasting, the authors explain.

Dr. Hany S. Bedair and colleagues from the University of Pittsburgh Medical Center, Pennsylvania, investigated the effects of systemic delivery of losartan on muscle regeneration and fibrosis in a laceration model of normal skeletal muscle injury in mice.

Three to five weeks after acute muscle injury, there were significantly more regenerating myofibers in mice treated with the ARB than in control mice, the researchers found, and there was significantly less skeletal muscle fibrosis within the zone of injury in ARB mice than in control mice.

The use of losartan "has been demonstrated in this study to enhance muscle healing ... in doses below those required for any antihypertensive effect," the investigators conclude.

"These findings may represent the most clinically appealing and potentially far-reaching antifibrotic therapy developed so far for otherwise healthy individuals who have sustained acute skeletal muscle injuries," they add.
